Highlights •We efficaciously obtained P. graminea transformants harboring silenced types of Pgmiox.•Pgmiox is required for response to osmosensitivity and fungicide tolerance of P. graminea.•Pgmiox is not only related to cell wall integrity, but also necessary for the pathogenicity in P. graminea.
