Abstract
For screening methanol and the metabolite formic acid in animal tissues, head space gas chromatography mass spectrometry (HS-GC/MS) analytical methods have been developed. Adult rats were toxic by vapor inhalation. Each sample was dispersed in benzyl alcohol and methanol (1% sulfuric acid) to determine the methanol and formic acid content of urine, serum, skin and lung tissue. Formic acid was derivatized with methyl formate in acidic conditions and used for analysis. Acetonitrile was used as the internal standard for methanol and formic acid analysis. The linearity of methanol and formic acid was both above 0.99. The methanol detection of limit (LOD) of the solid matrix was 0.04 μl/L and formic acid was 0.01 μl/L. Methanol limit of quantitation (LOQ) was 0.13 μl/L and formic acid was 0.02 μl/L. The methanol LOD of the liquid matrix was 0.07 μl/L and formic acid was 0.01 μl/L. The methanol LOQ was 0.21 μl/L and formic acid was 0.02 μl/L. The precision of the coefficient of variation (CV) was less than 15%. Accuracy and recovery were in the range of 80-120%. Identification was performed using GC/MS scan mode and selected ion monitoring (SIM) mode.
