Abstract
The effects of Cudrania tricuspidata root extract (CTE) on alcohol metabolism and to help maintaining blood homeostasis in rats treated with 10 ml/kg alcohol were investigated. CTE (500 mg/kg and 750 mg/kg) was treated after 30 minutes of alcohol ingestion (10ml/kg) in rats. Blood collection was implemented from the tail veins of the rats after 1, 3, 5 hours post alcohol treatment. The sobering drink (a commercial anti-hangover beverage) was used as a positive control. Consumption of CTE (500 mg/kg and 750 mg/kg) decreased serum ethanol concentration by increasing and maintaining both alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) enzyme activity levels in blood and liver. Also, CTE led to recovery from imbalances in blood gas levels, by regulating carbon dioxide (CO2). The level of pH and bicarbonate (HCO3-) were changed by regulation of CO2. Also, CTE reduced lactic acid levels increased by alcohol ingestion. Hepatoprotective effects of CTE (10-200 μg/mL) in 5% alcohol-treated Huh-7 hepatocarcinoma cell line were investigated using the MTT assay. In conclusion, CTE exerted an anti-hangover effect than a commercial anti-hangover drink and CTE can influence maintain blood homeostasis. Simultaneously CTE exhibited preventive effect of ethanol-induced hepatotoxicity. Therefore, we recommend that CTE can be a novel anti-hangover natural product agent for prevention or treatment of symptoms caused by excessive alcohol consumption.
