Abstract
This study identified the effect of Bacillus subtilis PL-2, which is an effective microorganism used for the preparation of traditional Korean fermented food, on the mechanism of the inflammatory reaction in mast cell by classifying the microorganism according to molecular weight into types with over 30 kDa, between 10~30 kDa, and under 10 kDa.The histamine release, pro-inflammatory cytokine(TNF-α, IL-8 and IL-6) secretion and mRNA trascription were suppressed according to concentration of treatment with the extract under 10 kDa of the Bacillus subtilis PL-2 extracts in mast cells. In order to confirm the molecular mechanism for the suppression of the inflammatory reaction by Bacillus subtilis extracts of under 10 kDa, the level of protein expression in relation to the NF-κB and MAPK pathway, which is a signal delivery system, was confirmed. As a result of confirming the effect of the Bacillus subtilis PL-2 extracts of under 10 kDa on NF-κB signal delivery, the phosphorylation of lκB was inhibited, which led to the suppression of the inflow of NF-κB molecules into the nuclei. As shown in the result of the confirmation of the phosphorylation of ERK, p38 and JNK, which have been known to play a critical role in triggering the inflammatory reaction in the MAPK pathway, the phosphorylation of ERK and p38 was confirmed as likely to decrease remarkably along with increasing concentration of PL-2 extract treatment.Therefore, the Bacillus subtilis PL-2 strain extract under 10 kDa was supposed to be effective in suppressing the allergic inflammatory reaction.
