Abstract
Periodontiti has been associated with the presence of a sub-gingival poly-microbial bio?lm containing T. denticola and P. gingivalis. In this study, we have investigated the co-culture condition and the potential synergistic virulence factors of T. denticola and P. gingivalis when in their co-culture. We identified differentially protein expression of T. denticola and P. gingivalis in single- and co-culture using the two-dimensional gel electrophoresis and MALDI-TOF Mass spectrometry analysis. Among eleven proteins,eight proteins were up-regulated while three proteins were down-regulated compare the single-culture to the co-culture. Among them, the stress response factors, metabolic factors were significantly up-regulated indicating that T. denticola becomes potentially more pathogenic in the presence of P. gingivalis. The differential RNA expressions of the related T. denticola genes were also confirmed by RT-PCR. These data revealed that T. denticola has a stronger viability and becomes more resistant for stress response and more potentially pathogenic virulence when in presence with P. gingivalis.
