Effect of plasmalogen on LPS-induced ICAM-1 expression in human gingival fibroblasts

Abstract
Periodontal disease is an inflammatory condition caused mainly by infection with periodontopathic bacteria. Plasmalogen (Pls) is glycerophospholipid that is one of the components of cell membranes and possesses various biological functions. We investigated the effect of Pls on intercellular adhesion molecule-1 (ICAM-1) expression in human gingival fibroblasts (HGF) stimulated with lipopolysaccharide (LPS) derived from Porphyromonas gingivalis (Pg). ICAM-1 expression was analyzed using real time PCR and flow cytometry. Pls decreased Pg-LPS-induced ICAM-1 gene expression in a dose-dependent manner in HGF. It was also revealed by flow cytometry that the addition of Pls decreased the cell surface expression of ICAM-1 in Pg-LPS-stimulated HGF. The results demonstrated that Pls suppressed Pg-LPS-induced ICAM-1 expression in HGF at the gene and protein levels. Our findings suggest that Pls may inhibit inflammatory reactions through the reduction of ICAM-1 expression in periodontal disease. (J Osaka Dent Univ 2022; 56: 13-17)
