Abstract
Skeletal muscle has excellent characteristics of high flexibility and efficiency for energy conversion and it has the possibility of a bio-actuator. The skeletal muscle cells were cultured by applying mechanical stimulation in the dishes. We evaluated the effect of this cultivation method and applied the cultured skeletal muscle to the bio-actuator. MethodCollagen gel was used as a scaffold of C2C12 cells and the cells were embedded. After 2 days in culture, the growth medium (GM) was shifted to the differentiation medium (DM), and the scaffold of C2C12 cells were cultured for total 23 days. After 10 days in culture, the skeletal muscle cells were applied mechanical stimulation in the dishes using multi mechanical stimulation. The contractile forces were measured and the results of HE stain were observed.ResultsThe results of the contractile forces of the skeletal muscles showed that applying mechanical stimulation in one direction was important for cultivation of the skeletal muscles. The effect of this cultivation method was evaluated and the myotubes grown in one direction were investigated in the results of HE stain cross sectional view.
