Abstract
The effect of endotoxin on bile flow and Na+-K+-ATPase activity in the liver cell membrane was examined. With an experimental model of intrahepatic cholestasis by use of TLCA, furthermore, circumstances surrounding the onset of endotoxinemia and the effect of change the reticuloendothelial function on such circumstances were analyzed. The administration endotoxin significantly reduced bile flow and Na+-K+-ATPase in liver cell membrane activity, which change was correlated to the endotoxin dose. The administration of TLCA turned Limulus test positive. When zymosan, that stimulates the reticuloendothelial system, was administered in conjunction with TLCA, Limulus test tended to be negative, and decreases in bile flow and liver cell membrane Na+-K+-ATPase activity were lessened. From the above findings, experimental intrahepatic cholestasis due to TLCA was considered most likely to be intensified by endotoxin, and further studies seemed necessary concerning changes in the lipid composition in the liver cell membrane.
