Abstract
The microscopic pK(sub a) values of thioredoxin (Trx) in Escherichia coli labeling and site-directed mutagenesis. Results obtained revealed that the difficulty in determining pK(sub a) values in reduced Trx resulted from the location of three titrating groups in the active site. The electrostatic interactions of the enzyme and the alpha-helix dipole had an opposing effects on the titration of Cys32.
