X-ray diffraction analysis of cytochrome P450 2B4 reconstituted into liposomes

Abstract
The depth of insertion of the microsomal cytochrome P450 protein into a phospholipid bilayer was measured by generating uniform proteoliposomes with P450 proteins in their bilayer, sedimenting them into stacked planar bilayers and exposing them to low-angle X-ray diffraction. The resultant one-dimensional unit-cell data indicated how deep the protein is in the intermembrane area. The distribution of lipids and proteins along the width of the membrane was determined by the electron density profile.
