Involvement of the GroE chaperonins in the nickel-dependent anaerobic biosynthesis of NiFe-hydrogenases of Escherichia coli

Abstract
The Escherichia coli chaperonins, GroES and GroEL, have a role in the biosynthesis of a functional Ni-Fe hydrogenase isoenzyme HYD3 through the incorporation of nickel. The GroES and GroEL thermosensitive mutant strains show a 60% reduction in total hydrogenase activity. A temperature shift during groE mutant growth inhibits HYD1 activity and allows accumulation of HYD3 precursor. However, processing of HYD2 and its activity remain unaffected. The nickel uptake mutant, nik, also shows a reduction in hydrogenase activity and an accumulation of precursors of HYD2 and HYD3.
