Abstract
Mutations in the T7 RNA polymerase initial transcription complex reveal that His784 establishes an H-bond with the 2-amino group of the initiating guanosine triphosphate and the 3'-guanosine of the transcript. Data suggest that this interaction directs both initiation with guanosines and retains G-terminated short RNAs within the complex.
