Abstract
Representational difference analysis (RDA), where human tumor cell DNA drives hybridization against normal DNA, helps detect genomic regions showing homozygous depletion in cultured cancer cells. Conversely, RDA also involves driving of hybridization by normal DNA against tumor cell DNA, isolating probes that detect amplification in tumors. Such bi-operational RDA performed on tumor DNAs isolates a probe that identifies frequent homozygous loss in colon cancer cell lines and localized to chromosome 3P.
