Abstract
A detailed, multistep purification procedure to characterize the components of active human gamma-secretase complex is reported together with the description of factors that modulate the production of amyloid beta-protein (A beta) by gamma-secretase. Mass spectrometry revealed heterodimeric presenilin (PS) composed of its N- and C-terminal fragments (PS-NTF/CTF), a mature glycosylated form of nicastrin (NCT), Aph-1 and Pen-2, as components of active gamma-secretase.
