Abstract
The ability of the Escherichia coli cosmid library of Haemophilus somnus DNA to bind hemin (Hmb+) was analyzed to characterize the mechanisms of heme uptake in Haemophilus somnus. Transposon mutagenesis of the HindIII fragment of Haemophilus somnus DNA that conferred the Hmb+ phenotype in the host strain indicated the role of the orf7 gene in encoding a protein that was associated with the Hmb+ phenotype. Furthermore, the orf7 gene product also significant levels of lysozyme activity which mediated the uptake of heme.
