Abstract
Kinetic studies of the interaction of the nuclear GTP-binding protein Ran with two types of its regulatory proteins, RCC1 and RanGAP1, reveals that presence of these regulatory proteins significantly increases the guanine nucleotide dissociation and GTPase reactions of Ran. Use of fluorescent, radiolabeled and unlabeled guanine nucleotides and the intrinsic tryptophan fluorescence of Ran helps follow the kinetics. Experimental studies with Ran mutants T24N and Q69L help understand these interaction.
