Abstract
Preservation of full fibrinolytic activity under reduced clearance is achieved by a combination of mutations at distinct sites on tissue plasminogen activator (tPA). An additional mutation, N117Q(N), is applied to maintain fibrinolytic activity. A more potent variant (TNK-tPA) exhibiting advantages for clinically-induced thrombolysis is produced.
