Abstract
A dose–response curve to X-Rays by Cytokinesis – Block Micronucleus (CMBN) assay was realised for medical dose estimation by the irradiation of blood samples obtained from six healthy donors. The irradiation was performed at room temperature using two X-ray beams at 6MV voltage, the dose rate of 240 cGy / min and the parameters time, monitor units (MU) were chosen using the Varian Eclipse 11 treatment plan system. Two cell culture were initiated for each dose - from 0 to 4 Gy, the final each dose result being reported at 1000 binucleated lymphocytes. For each irradiation dose were analyzed manually 12 000 binucleated lymphocytes. Results were fit with linear-quadratic dose–effect model. Dose Estimate program for fitting calibration curves was used; variations in the background frequency, age and sex categories were also observed, in accordance with results of the cited literature and recommendations for each laboratory to perform in vitro its own dose-response calibration curve.
