Abstract
目的 探讨钙离子激活氯离子通道蛋白7(ANO7)在前列腺癌核因子-κB(NF-κB)信号通路的表达及其临床意义.方法 利用Tribbles同源蛋白1(TRIB1)过表达前列腺癌细胞株构建NF-κB信号通路激活细胞和动物模型,NF-κB信号通路抑制剂构建NF-κB信号通路抑制细胞模型.用蛋白质印迹法(Western blot)、实时定量聚合酶链反应(Real-time PCR)和免疫组织化学(IHC)分析前列腺癌中NF-κB信号通路对ANO7表达的调控影响,分析临床数据中ANO7表达与前列腺癌临床病理特征之间的关系.结果 在NF-κB信号通路激活细胞和动物模型中可见ANO7表达上调;在NF-κB信号通路抑制细胞模型中可见ANO7表达下调.组织芯片中ANO7在肿瘤组织和癌旁组织中高表达率分别为45.8%、85.7%,差异有统计学意义(x2=7.258,P<0.01).ANO7表达与Gleason评分、病理分级显著相关(x2=19.797,19.797,P<0.01),癌症基因组图谱(TCGA)与泰勒(Taylor)数据库得到相似的结果.运用Kaplan-Meier分析发现在Taylor数据库中ANO7与患者的预后明显相关(P<0.01).最后采用Cox回归模型进行多因素综合分析发现在影响前列腺癌预后的队列中,ANO7 (P <0.05)、Gleason评分(P<0.01)和病理分级(P<0.01)都是前列腺癌的独立预测指标.结论 NF-κB信号通路调控前列腺癌ANO7表达,且ANO7的表达与前列腺癌明显相关.
Objective To study the Expression of anoctamin 7 (ANO7) in the nuclear factor kappa B (NF-κB) signal pathway and its clinical significance in prostate cancer.Methods NF-κB signal pathway activated cell and animal model were constructed using tribbles homolog1 (TRIB1) overexpressed prostate cancer cell line.NF-κB signal pathway inhibited cell model was constructed using NF-κB signal pathway inhibitor.Western blotting and real-time quantitative polymerase chain reaction (Real-time PCR) and immunohistochemistry (IHC) were used to analyze the effect of NF-κB signal pathway on the regulation of ANO7 expression and the relationship between ANO7 expression and clinicopathological features in prostate cancer.Results ANO7 expression was up-regulated in the NF-κB signal pathway-activated cell and animal models.ANO7 expression was down-regulated in the NF-κB signal pathway-inhibited cell models.The high expression level of ANO7 was 45.8% in tumor tissues compared with 85.7% in adjacent non-cancerous tissues,with statistical significance (x2 =7.258,P < 0.01).The expression of ANO7 was significantly correlated with Gleason score and pathological grade (x2 =19.797,19.797,P < 0.01).Similar results were obtained from The Cancer Genome Atlas (TCGA) and the Taylor database.Kaplan-Meier analysis found that ANO7 was significantly associated with the patients' prognosis in the Taylor database (P < 0.01).Multivariate analysis found that ANO7 (P < 0.05),Gleason score (P < 0.01) and pathological grade (P < 0.01) were independent predictors of prostate cancer using the Cox regression model in the cohort affecting prostate cancer prognosis.Conclusion NF-κB signal pathway regulates ANO7 expression in prostate cancer and the expression of ANO7 is closely related to prostate cancer.
