Abstract
目的 观察麝香乌龙丸对兔膝骨关节炎软骨细胞凋亡和凋亡调控基因bcl-2及其抑制基因bax的影响,探讨麝香乌龙丸对关节软骨的保护作用是否与抑制细胞凋亡与调节bcl-2及bax的表达有关.方法 选用健康日本大耳白兔30只,随机分成正常对照组、造模组、麝香乌龙丸组.用1.6%木瓜蛋白酶膝关节腔内注射,建立兔膝骨关节炎模型, 4周后处死全部动物,取右膝关节胫骨内侧髁软骨,软骨组织切片用TUNEL法检测软骨细胞凋亡,免疫组化法检测关节软骨中bcl-2、bax的水平.结果 麝香乌龙丸组细胞凋亡率明显低于造模组(P<0.05),bcl-2在麝香乌龙丸组中的表达高于造模组(P<0.05),bax的表达低于造模组(P<0.05).结论 麝香乌龙丸通过上调bcl-2的表达,下调bax的表达,达到抑制兔实验性膝骨关节炎软骨中软骨细胞的过度凋亡,这可能是该药治疗骨性关节炎机制之一.
Objective To observe the effect of Shexiang-wulong pill on the rabbit knee osteoarthritis( KOA) chondrocyte apopto-sis, apoptosis regulatory genes bcl - 2 and its impact suppressor gene bax. To explore the relationship of Shexiang - wulong pill on the protective effect of articular cartilage with the inhibition of apoptosis and regulation bcl - 2 and bax expression. Methods 30 healthy male rabbits were randomly divided into three groups; A. normal control group, B. model group,C. Shexiang - wulong pill group. Make KOA models by injecting papain to the right knees of rabbits, after 4 weeks all animals were put to death. Then take the right knee medial tibial condyle cartilage. Cartilage tissue with cartilage TUNEL assay of apoptosis, detected by immunohisto-chemistry of articular cartilage in the bcl - 2, bax level. Results The rate of apoptosis of the group C was significantly lower than of B(P<0.01), the expression of bcl-2 in C was higher than that in B(P<0.01), the expression of bax in,the C was lower than in B ( P<0.01). Conclusion It plays a role by upregulating the expression of bcl - 2, reducing the expression of bax to inhibit the excessive apoptosis of chondrocytes on the rabbit KOA, this may be one mechanism of Shexiang -wulong pill on the treatment of KOA.
