Abstract
目的 探讨三结构域蛋白59(TRIM59)、真核翻译起始因子5A2(EIF5A2)在非小细胞肺癌组织中的表达及对转移分化的预估价值.方法 选取非小细胞肺癌组织80 例、正常肺组织 80 例,qRT-PCR法、免疫组化法检测 2 种组织中TRIM59、EIF5A2 mRNA与蛋白表达情况.收集肺癌患者临床资料,根据其是否出现转移分化进行分组:转移分化组、未转移分化组,分析其出现转移分化的影响因素.绘制ROC曲线分析TRIM59、EIF5A2 表达对肺癌患者转移分化的预估价值.结果 NSCLC组织中TRIM59、EIF5A2 mRNA表达水平高于正常组织,TRIM59、EIF5A2 蛋白高表达率高于正常组织(P<0.05).转移分化组与未转移分化组患者之间的TRIM59、EIF5A2 蛋白表达情况、肿瘤分化程度差异有统计学意义(P<0.05),TRIM59、EIF5A2 蛋白高表达和肿瘤低分化为NSCLC肿瘤转移分化的危险因素.NSCLC组织中TRIM59 联合EIF5A2 蛋白表达水平预估肿瘤转移分化的 AUC 以及特异度均明显高于单独检测.结论 TRIM59、EIF5A2 在非小细胞肺癌组织中的表达水平异常升高,是非小细胞肺癌转移分化的影响因素之一,且对其转移分化有一定预估价值.
Objective To investigate the expressions of three-domain protein 59(TRIM59)and eukaryotic translation initiation factor 5A2(EIF5A2)in non-small cell lung cancer tissues and their predictive value for metastatic differentiation.Methods 80 cases of non-small cell lung cancer tissues(lung cancer group)and 80 cases of normal lung cancer tissues(nor-mal group)were selected,and the mRNA and protein expressions of TRIM59 and EIF5A2 in the 2 kinds of tissues were detected by qRT-PCR and immunohistochemistry.The clinical data of the lung cancer patients were collected and divided into groups ac-cording to whether they had metastatic differentiation:metastatic differentiation group and non-metastatic differentiation group,and the influencing factors of metastatic differentiation were analyzed.The ROC curve was drawn to analyze the predictive value of TRIM59 and EIF5A2 expression on the metastasis and differentiation of lung cancer patients.Results The mRNA expression levels of TRIM59 and EIF5A2 in NSCLC tissues were higher than those in normal tissues,and the proportion of those with high protein expression of TRIM59 and EIF5A2 was higher than that in normal tissues(P<0.05).There were significant differences in the expression of TRIM59 and EIF5A2 proteins and the degree of tumor differentiation between the metastatic differentiation group and the non-metastatic differentiation group(P<0.05).risk factors.The AUC and specificity of TRIM59 combined with EIF5A2 protein expression in NSCLC tissues for predicting tumor metastasis and differentiation were significantly higher than those of single detection.Conclusion The expression levels of TRIM59 and EIF5A2 in non-small cell lung cancer tissues are ab-normally increased,which are one of the influencing factors of the metastasis and differentiation of non-small cell lung cancer,and have certain predictive value for the metastasis and differentiation of non-small cell lung cancer.
