Abstract
目的:探讨ADC值在下腔静脉梗阻型布-加综合征(BCS)大鼠模型肝脏氧化应激损伤中的应用价值.方法:选择健康雄性SD大鼠135只,随机分为对照组(15只,常规清洁饲养)、模型组(60只,结扎大鼠肝后段下腔静脉)和假手术组(60只,不结扎下腔静脉).模型组及假手术组各分为4个亚组(1、4、8、12周组,每亚组15只).观察各组在不同时间点肝脏ADC值的变化,以各组肝脏HE染色情况,检测各组丙二醛(MDA)、超氧化物歧化酶(SOD)的变化.结果:模型组ADC值呈现先下降后升高趋势(第4周最低),始终低于对照组及假手术组,差异均有统计学意义(均P<0.05).模型组术后MDA先升高后下降趋势,始终高于对照组及假手术组(均P<0.05);SOD呈先下降后升高趋势,始终低于对照组及假手术组(均P<0.05).模型组ADC值与MDA呈负相关,与SOD呈正相关,SOD与MDA呈负相关(均P<0.05).结论:ADC值与BCS的肝脏淤血缺氧后的氧化应激损伤存在一定的相关性,可作为动态评估BCS肝脏淤血缺氧损伤的新的检查手段.
Objective:To explore the application value of ADC value in oxidative stress injury of the liver in a rat model of Budd-Chiari syndrome.Methods:135 experimental SD rats were randomly divided into a control group(15 rats,routine clean feeding),model group(60 rats,ligation of the posterior segment of the inferior vena cava)and sham-operation group(60 rats,no ligation of the inferior vena cava).The model group and sham-operation group were divided into four subgroups(1-,4-,8-,12-week group,15 rats in each subgroup).The changes of ADC values in the liver at different time points were observed.HE staining of the liver in each group was observed,and the changes in malonaldehyde(MDA)and superoxide dismutase(SOD)were detected.Results:The ADC value of the model group showed a trend of decreasing first and then increasing(the lowest in the fourth week),which was always lower than that of the control group and the sham-operation group,the overall difference was statistically significant(all P<0.05).After operation,the MDA value of the model group first increased and then decreased and was always higher than that of the control group and the sham-operation group,while the SOD value first decreased and then increased and was always lower than that of the control group and the sham-operation group(all P<0.05).The ADC value of the model group was negatively correlated with MDA value and positively correlated with SOD value,and SOD value was negatively correlated with MDA value(all P<0.05).Conclusions:The ADC value has a certain correlation with oxidative stress injury in Budd-Chiari syndrome after hepatic congestion and hypoxia,and it can be used as a new method to dynamically evaluate hypoxic injury in Budd-Chiari syndrome.
