双丹明目胶囊对糖尿病视网膜病变大鼠血糖血脂及血液流变学的影响 / Effects of Shuangdan Mingmu Capsule on Blood Glucose, Blood Lipid and Blood Rheology in Diabetic Retinopathy Rats

Abstract
目的 观察双丹明目胶囊对糖尿病视网膜病变(diabetic retinopathy, DR)大鼠血糖血脂及血液流变学的影响,探讨其可能的治疗机制.方法 SD大鼠经一次性尾静脉注射STZ(50 mg/kg)诱导DR模型,随机分为模型组,双丹明目胶囊高、中、低组,导升明组,同时以正常大鼠为对照.连续干预30 d后,生化法检测大鼠血糖、血脂水平,全自动血液流变快测仪检测大鼠全血粘度(WBV)、血浆粘度(PV)、红细胞沉降率(ESR)、红细胞压积(HCT)、纤维蛋白原(FIB)等血液流变指标.结果 与正常组比较,模型组大鼠血糖、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)水平均显著升高(P<0.01),高密度脂蛋白(HDL-C)水平显著下降(P<0.01),WBV、PV、ESR、HCT、FIB水平均显著升高(P<0.01或P<0.05);双丹明目胶囊干预后,模型大鼠血糖、TC、TG、LDL-C水平均下降(P<0.05),HDL-C上升(P<0.05),WBV、PV、ESR、HCT、FIB水平均下降(P<0.05).结论 双丹明目胶囊治疗DR的机制可能与有效改善DR大鼠血糖血脂紊乱、血液流变异常状态有关.
Objective To observe the effects of Shuangdan Mingmu capsule on blood glucose, blood lipid and blood rheology, and to explore its possible mechanism of treatment. Methods SD rats were injected with STZ (50 mg / kg) into the tail vein to induce diabetic model with retinopathy. The rats were randomly divided into model group, high dose, medium dose and low dose of Shuangdan Mingmu capsule groups and positive drug group, normal rats as controls. After continuous administration for 30 days, the blood glucose and blood lipid of rats were measured by biochemical method. The hemorrheological indexes were tested such as whole blood viscosity (WBV), plasma viscosity (PV), erythrocyte sedimentation rate (ESR), hematocrit (HCT) and fibrinogen (FIB). Results Compared with the normal group, the levels of blood glucose, total cholesterol (TC), triglyceride (TG) and low density lipoprotein (LDL-C) in model group increased significantly (P<0.01) and high-density lipoprotein (HDL-C) levels decreased significantly (P<0.01). The levels of WBV, PV, ESR, HCT, FIB increased obviously (P<0.01 or P<0.05). After administration of Shuangdan Mingmu capsule, blood glucose, TC, TG, LDL-C levels of model rats decreased (P<0.05), HDL-C increased (P<0.05), the levels of WBV, PV, ESR, HCT, FIB declined (P<0.05). Conclusion The mechanism of Shuangdan Mingmu capsule in the treatment of DR might be related to the effective improvement of blood glucose and lipid disorders, and abnormal blood rheology in DR rats.
