Abstract
目的:观察粒细胞集落刺激因子(G-CSF)对动脉粥样硬化形成的影响,并对其机制进行初步探讨,为临床提供参考。方法将30只新西兰雄性健康大白兔随机分为对照组6只,高脂喂养组、普通及高脂喂养+G-CSF组各8只,4组动物每周采血,监测总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)等各项血脂指标动态变化。取材后用苏丹Ⅲ染色观察脂肪斑块面积,苏木精-伊红(HE)染色观察内膜厚度等形态学变化。免疫组化观察基质金属蛋白酶(MMP)9及其抑制物(TIMP-2)的表达。并用Leica QwinV3图像分析系统计算阳性率等相关指标。扫描电镜观察内皮及脂肪细胞的组间差异。结果高脂喂养能使TC、LDL等血脂水平上升,6周后高峰,与对照相比,P<0.05,但8周后就趋于稳定。加G-CSF使血脂高峰前移,使TC、LDL等提前2周就与对照组出现统计差异(P<0.05),并有双峰样改变,但对TG同样无影响。普通喂养+G-CSF能使血脂有所波动,但尚不足以达到显著差异。病理分析发现G-CSF能使斑块面积、内膜厚度、MMP/TIMP表达均增加,但后二者并未达到统计学差异(P>0.05)。扫描电镜支持上述发现。结论 G-CSF对血脂存在一定影响,且以TC、LDL为主,对TG作用不大。因此它可使动脉粥样硬化恶化,具体表现为斑块面积的扩大,内膜厚度增加,机制可能与其对基质的影响有关,但尚需进步研究证实。
Objective Inorder to give reference for clinic application,we investigated the influence on atherosclerosis of hyperglycemia rabbit by granulocyto colony-stimulating factor(G-CSF). Methods Thirty New Zeeland healthy male rabbits were divided into 4 groups randomly,whose blood was drew from the ear vein weekly to test the change of total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL),low density lipoprotein (LDL). After sacrifice,the aorta thoracicalis was taken to make the SudaneseⅢfat dye,conventional hematoxylin and eosin(HE)dye as well as scanning electron microscope to observate the form change of endothelial cells,gruel type mottling and fat foam. Meanwhile immunohisto chemical IHC of matrix metallopeptidase 9(MMP9) and tissue inhibitor of metalloproteinase 2(TIMP2) was used to explorate the maitrix,and many datas of pathology were calculated by Leica QwinV3 image analysis system. Results Fed with high fat food,the level of blood fat such as TC,LDL and so on can rise to peak in 6 weeks,then go down to stable platform period after 8 week. G-CSF can make the fat curve appear double-peak,and the statistics difference of TC,LDL arrives 2 weeks earlier,contrast to the control group(P<0.05). But it had no effect on TG. Adding G-CSF to ordinary fed group can make those blood fat have some fluctuation but can’t get the significant deviation. The analysis by pathology also found G-CSF can increase the plaque area,endomembrane thickness,and the express of MMP9/TIMP2. However only the former index won statistics significance between the 2 groups fed with fat. The scans electron microscope found the fragments of endothelial cell increases obviously when atherosclerosis appeared,shrinks in shape, so the same point was surported. Conclusion G-CSF has certain effect on the endothelial cell shape,and stimulate the blood fats level,primarily TC,LDL. For this reason it can aggravate atherosclerosis. The mechanism maybe related to maitrix but need further verification.
