血清外泌体中lncRNA SOX21-AS1的表达在宫颈癌诊断中的临床价值 / The clinical significance of serum exosome-bsed evaluation of lncRNA SOX21-AS1 levels for cervical cancer diagnosis

Abstract
目的 检测血清外泌体中SOX21-AS1的表达水平,评价其在宫颈癌中的诊断价值.方法 选取130例宫颈癌患者作为观察组,包括宫颈鳞癌患者98例(高分化鳞癌30例,中分化鳞癌41例,低分化鳞癌27例)、宫颈腺癌26例、腺鳞癌6例.选取同期健康体检女性50例作为对照组.采用低温差速离心法提取外泌体,qRT-PCR法检测所有患者血清及血清外泌体SOX21-AS1的水平.结果 宫颈癌患者血清中SOX21-AS1、外泌体中SOX21-AS1水平,均较健康对照组显著升高,差异有统计学意义(P<0.05);宫颈癌患者血清中与血清外泌体的SOX21-AS1水平,差异无统计学意义(P>0.05).血清外泌体中SOX21-AS1水平的ROC预测AUC(0.9543),高于血清中SOX21-AS1水平的ROC预测AUC(0.9267),差异有统计学意义(P<0.05).结论 血清外泌体中的SOX21-AS1水平可以作为宫颈癌临床诊断的生物标志物.
Objective To determine the degree of SOX21-AS1 expression within serum exosomes and ascertain its diagnostic potential for cervical cancer.Methods The observation group comprised 130 patients diagnosed with cervical cancer,out of which 98 were diagnosed with cervical squamous cell carcinoma(30 patients with grade Ⅰ highly differentiated squamous cell carcinoma,41 patients with grade Ⅱ moderately differentiated squamous cell carcinoma,and 27 patients with grade Ⅲpoorly differentiated squamous cell carcinoma),26 were diagnosed with cervical adenocarcinoma,and 6 were diagnosed with adeno-squamous cell carcinoma.Fifty healthy women were selected as the control group.The exosomes were extracted through low-temperature differential centrifugation,and the levels of serum SOX21-AS1 were detected in all patients through qRT-PCR.Results The levels of SOX21-AS1 in the serum and exosomes of cervical cancer patients were significantly higher than those in the healthy control group,with a statistically significant difference(P<0.05).No significant difference was found in the level of SOX21-AS1 between serum and serum exosomes in patients with cervical cancer(P>0.05).The area under the receiver operating characteristic(ROC)curve for SOX21-AS1 levels in serum exosomes(0.9543)was greater than that for serum SOX21-AS1 levels(0.9267),with a statistically significant difference(P<0.05).Conclusions The concentration of SOX21-AS1 in exosomes found in serum has the potential to serve as a reliable biomarker for the clinical diagnosis of cervical cancer.
