Abstract
The species identification for the products of ivory specimens by the analysis system of multiplex-PCR (polymerase chain reaction) was established in this study. The primers specific for the Elephasidae of Cytochrome b (Cytb), Cytochrome c Oxidase subunit I (COI), 16S rRNA, and 12S rRNA on mitochondrial genome were designed and the multiplex PCR for three different size fragments were developed. The standard elephant DNA were collected from two African elephants and three Asian elephants for the reaction condition optimization testing and system establishment. The three different size PCR products from these standard DNA can be amplified and their DNA sequences can be successfully obtained. This system was applied on testing of 10 suspected ivory fragments. The results showed that 7 to 9 specimens of each gene can be amplified successfully with at least one size fragment of PCR products. The species with the highest similarity of the DNA sequences from these PCR products were Loxodonta africana or Loxodonta cyclotis after comparing with the sequences registered in GenBank by the basic local alignment search tool (BLAST program). The system established in this study can effectively achieve the goal of shortening the identification time of ivory products and reducing the loss of specimens.
