Abstract
In this study, Alavattam et al. investigate the dynamic regulation of H3K9me3 in male meiosis and identify the germline-specific protein ATF7IP2 (MCAF2) as a novel, critical regulator of H3K9 methylation and spermatogenic heterochromatin organization. They show that ATF7IP2 spatially directs histone methyltransferase SETDB1 to control the distribution of H3K9me3, thereby regulating heterochromatin organization essential for meiotic and autosomal gene regulation, meiotic sex chromosome inactivation, the expression of transposable elements, and male fertility.
