Abstract
We are interested in correlating the performance of fipronil in populations of cat fleas (Ctenocephalides felis) with potential cross-resistance conferred by point mutations in the Resistance to dieldrin gene, Rdl. Here we report the sequencing of exon 7 of the cat flea Rdl gene and the documentation of a putative resistance-associated mutation predicting the replacement of alanine302 with a serine. We describe two polymerase chain reaction (PCR) based diagnostics for the detection of this mutation. First, PCR mediated detection of a restriction endonuclease polymorphism (PCR REN) using a BsmAI site created by the resistance-associated mutation. Second, a TaqMan assay using allele specific fluorogenic probes and the TaqMan 5′ specific nuclease. We describe how such diagnostics can be used in the diagnosis of resistance in the field and laboratory. [Copyright &y& Elsevier]
