Abstract
1 Owing to clinical and histologic overlap, the differential for AFX often includes squamous cell carcinoma (SCC), melanoma, leiomyosarcoma, and cutaneous undifferentiated pleomorphic sarcoma (UPS). 4 Althoughwealso identified TP53 and CSMD3 mutations in all AFX, the other previously reported genes were not prevalent, although removal of the p-value threshold revealed NOTCH1 mutations in 3 of our 4 AFX samples. Many mutations seen in AFX and UPS are shared by cutaneouscancers.TP53ranksamongthemostcommonly mutatedgenesacrossvarioushumantumortypes, including cutaneous SCC. Although priorstudieshavesuggestedthemutationprofilecanaidin distinguishing AFX from other poorly differentiated neoplasms,thenumberofUV-inducedchangesmakesfindinga specific mutation for AFX versus UPS challenging.
