Abstract
The efficiency of AST is quantified by measuring the relative ratio of viable cells between the adherent and suspension cell populations using CellTiter Glo® 2.0 cell viability assay (Promega, G9241). Melanoma primary tumor and lung metastasis tissue dissociation GFP-positive B16F10 cells were isolated from the primary sites and lungs. Paired-end sequencing was performed using NovaSeq 5000/6000 S1 Rgt Kit v1.5 (200 cycles) on a NovaSeq600 (Illunmina) sequencer. scRNA-seq analysis of primary tumor cells and CTCs The Illumina outputs of human and mouse samples were mapped to the human reference genome (GRCh38) using Cell Ranger 6.0.0 (10X Genomics). Cell viability and ROS detection assay In vitro cell viability was assessed using the CellTiter Glo® 2.0 cell viability assay (Promega, G9241) and LIVE/DEAD® cell imaging kit (ThermoFisher, R37601) according to the manufacturer's protocols.
