Abstract
Objective: This study was designed to investigate the antimetastatic and immunomodulating effects of extracts of Ojeoksan on cancer cell.Methods: Cell viability of Ojeoksan was tested with colon 26-M3.1 carcinoma cells and splenocytes in vitro. Antimetastatic experiments were conducted in vivo mouse model by using colon 26-M3.1 carcinoma cell. To observe immunomodulating effects of Ojeoksan on peritoneal macrophages, we measured interleukin (IL)-6, IL-12, tumor necrosis factor-alpha (TNF-α) and monocyte chemotactic protein-1 (MCP-1). In addition expression of nitrite oxide induced by lipopolysaccharide (LPS) was measured after treating Ojeoksan. Cell cytotoxicity was tested with the macrophage stimulated with Ojeoksan. Metastasis was evaluated in mouse with adoptive transfer macrophage that stimulated with Ojeoksan. We evaluated the activation of interferon (IFN)-γ and natural killer (NK)-cell. And the effect of Ojeoksan on metastasis was measured without NK-cell using anti-asialo-monosialotetrahexosylganglioside (GM1) serum. Results: In vitro cytotoxicity analysis, the inhibitory concentration (IC)50 of the colon 26-M3.1 carcinoma cell was 900㎍/㎖. IC50 of the splenocyte with concanavallin A was 1030㎍/㎖. And IC50 of the splenocyte with LPS was 1030㎍/㎖. Ojeoksan did not show the tumor specific cytotoxicity. We found that the administration of Ojeoksan significantly inhibited tumor metastasis in vivo. Level of cytokines such as IL-6, IL-12, TNF-α and MCP-1 of Ojeoksan group were up reglulated in compared to the control group. The expression of nitrite oxide induced by lipopolysaccharide after treating Ojeoksan was down regulated in dose-dependent manner. The macrophage stimulated with Ojeoksan significantly inhibit the cancer cell at ratio of 20:1. Mouse with adoptive transfer macrophage that stimulated with Ojeoksan significantly inhibit metastasis of cancer. The activation of NK-cell was not significantly up regualted with Ojeoksan. The activation of IFN-γ was significantly up regualted in a group of 12.5:1. After the depletion of NK-cells by anti-asialo GM1 serum, the inhibitory effect of Ojeoksan significanty lasted on tumor metastasis.Conclusion: Ojeoksan appears to have considerable activity on the anti-metastasis by activation of immune system.
