TMS-19-Q is a new 16-membered macrolide antibiotic. In the course of clinical trial, the tablet designated as TMS-19-Q·O tablet formulated with usual ingredients was changed to TMS-19-Q·.GC tablet which was elaborated by formulating with glycin and citric acid to improve the absorption of the drug at any acidity of gastric juice.Plasma concentration and urinary excretion were compared after the administration of both tablets to 5 hospitalized patients with respiratory tract infection in crossover, and clinical trial was made in 7 patients with O-tab. and 8 with GC-tab. The results obtained were as follows:1. Maxima plasma concentrations of O-and GC-tab. at the doses of 400mg in fasted were observed 1.53μg/ml at 30min. and 4.3μg/ml at 30min. respectively. Four patients were aged over 65 and 3 patients showed hypoacidity. Urinary recovery up to 6 hours were 0.94% on O-tab. and 1.95% on GC-tab.2. Clinical efficacy rate of O-and GC-tab. was 50.0% and 71.4% respectively.3. No serious side effect was observed, 2 cases of slight elevation of S-GPT and S-GOT on O-tab. and 1 case of anorexia on GC-tab. were noted.These results suggest that TMS-19-Q·GC tab. would be useful for respiratory tract infections.
