Formation of multiple complexes between actinomycin D and a DNA hairpin: structural characterization by multinuclear NMR

Abstract
High-resolution NMR spectroscopic studies on solution complexes between the hairpin DNA oligonucleotide d(TCGCGTTTTCGCGA) and the anticancer drug actinomycin D (ActD) reveal that the binding mode of ActD to the DNA hairpin is similar to that of conventional double-stranded DNA. Two ActD-DNA complexes vary according to the orientation of the asymmetric chromophore-induced intercalation of the d(GC) step. NOESY spectra reveal the formation of 86 intermolecular contacts of which 27 contacts have interchangeable protons for each complex.
