Abstract
-We map DDX41 germline variation in 454,792 volunteers and define the risk of MDS/AML development associated with different variant types.-DDX41-mutant MDS/AML evolves differently to sporadic disease, but individuals at high risk often have somatic DDX41 mutations or a high MCV.
